Viruses can't replicate on their own so you have to do it surrounded by a cell culture. Look up "cell culture" and you will find lots of information as this has be done for quite a few years presently.
Nurse a cold, keep the kleenex.
Don't cultivate virus unless you are trained to do so. Viruses can be extremely dangerous.
